I don't really remember any game that made me laugh out hard...Do you?
Maybe it's a thing with the own games medium, they usually don't convey much hilarity. Of course they work a lot better when trying to get some tears from us, but what about another emotions.

Tim Schafer's games always have great funny moments, though my particular favorite is Psychonauts.

Nobi Nobi Boy and GTA4 multiplayer are the only examples I can think of, but I'm sure there are a few more. I'm not sure whether I've played a game that was intentionally funny and succeeded in making me laugh, though. Then again, I generally don't laugh much on my own, even when I find things funny.

Perhaps not the answer you were looking for: Any game with a good coop or multiplayer - Laughing at stuff like super smash bros in the sofa with friends or over ts/vent/whatever. Alone... hmm, laughing at bad ai

For me, there were definitely some laugh out loud moments in Dragon Age and Mass Effect, the banter between Morrigan and Alistair in particular.

Lucas Arts and more recently Telltale games are notorious for this kind of stuff. I played the Sam & Max new games a while ago and it was a great source of laughter.
Playing as a Malkavian in Vampire: The Masquerade - Bloodlines also has some hillarious moments.
More unintendedly (maybe, or maybe not), Bayonetta. With all the absurd situations and excessive poses, I can't help but laugh.

After two years of owning it, Little Big Planet online gameplay still makes me laugh out many times when playing with friends.
Watching them panic, try to survive then die a silly death, zoom off somewhere on a rocket at 0-100mph in a second or two. I think the animations and emotional faces of Sackboy help, though.

Recently Fable 3 and Deadly Premonition have made me laugh quite muchly. GrappleBoy also produced a number of laughs when I played it recently.
Really, games are great at making you laugh. You can do such ridiculous non-sequiteur stuff in them.

Ok...just remembered, I laughed hard while playing warioware smooth moves with some friends. Anyway, I think it's much easier to do multiplayer games funny, where the funny thing is watching your friends fail in humiliating ways (Like they say, all laughter is sadistic)

The dialogue in Starship Titanic often had me in stitches, the elevator robot in particular was great for 'answering' random questions. The game was as frustrating as it was hilarious, though. Psychonauts, most definitely. Myth: The Fallen Lords got some big laughs, usually with dorf grenade physics related hilarity.
It's funny, if you look back at old games, like when DOS was king, almost all games has elements of humour. Modern games tend to be so serious and dreary. I know there are plenty of exceptions, especially in the indie scene, but the mainstream seems to have largely abandoned laughter, which is odd considering how popular comedy is in television and film.
