well, looking at the trailer and screenshots I'm hoping those aren't really missing enemies but just something for trailer purposes or being wip or something. Because missing enemies would suggest butchered difficulty.
It was never the enemies that brought the pain. It was the puzzles.

I wonder if they'll include the "make yourself small!" puzzle to the revamp.

I've found that the easy way to reduce difficulty in hard puzzle games without drastically changing everything (and probably what Nicalis will do) would be adding a fortune cookie mechanic where the player can spend a small amount of money to get a hint on what to do next.

Yeah, I can't imagine a game like this selling well. Metroid and Kid Icarus are easier.

I say WiiWare is taking away half the fun by removing the MSX references (the roms, the graphics). It LOOKS cool though... I would get, but I'm not that into games that are mostly hard cuz of puzzles.

I was looking forward to this, I just hope Nicalis doesn't manage to screw it up at the last minute. The last thing I want is music glitches on La-Mulana of all games. Fortunately, Nigoro seems to be in control of the game's development for the most part, though I'm still weary of Nicalis after the reported problems with CS.

They're clearly making some significant changes, considering that the lantern of time seems to have been removed entirely.

I seriously doubt it's going to have as high an entry barrier as the freeware version. On the other hand, I'm interested in seeing what happens to Confusion Gate, which was basically a collection of evil-bastard-puzzles in the original.

I think on the dev blog Nigoro mentioned some sort of hint/walkthrough system to accompany the game.
