Yes, the game should run fine in Vista. If you're running a Vista PC that supports Aero Glass, then you have definitely met the hardware requirements.. but it can be tricky to get all the required XNA software installed.
Anyways, I'll be happy to help everyone in the beta testing program for Owlboy related xna problems, so if you want to keep any issues about it inside the tester's program I can help out.
