Sometimes it's even appropriate and the game was attempting to be humorous.

also pretty much any game has a funny moment here and there that i laughed at, even if the game wasn't specifically trying to make me laugh. it'd be impossible to name all of those, but here's one example of a funny opening to a game (the game itself was one of my favorite ps1 games, but the intro is hilarious):

Yes, he just writes two paragraphs about it.
I was just playing Mount&Blade and had a jolly good time shooting people on horseback with my crossbow as they went over hills so that they went flying a fair few metres. It was a scream.

Leisure Suit Larry 6: Shape Up or Slip Out! Is probably the only one that has made me laugh, others can make me chuckle, but only Larry 6 made me LOL a couple of times.

I laugh often at games that aren't supposed to be laughed at. Like, the take themselves so seriously at points it's hilarious. Examples include Bionic Commando, Darksiders, Resi 4.
Oh man, Resi 4 made me laugh so hard, especially

Dragon Quest IX has made me giggle several times, mostly due to its absurdly abundant use of alliterations and puns. When you're fighting beasts like Sacksquatch (a living flour bag with teeth), it's hard not to laugh.

Maniac Mansion, Sumotori Dreams (that one practically had me in tears I was laughing so hard), Octodad, Earthbound, any game that allows your token-griefer-friend to go to town (like the Wii Super Mario Bros.)

A lot of the LucasArts point and click games are really funny in my opinion. Indiana Jones and the Fate of Atlantis has better lines in it than any of the movies, and Sam and Max Hit the Road is just really funny in a general sense. Planescape: Torment, Baldur's Gate II and Ultima VII, which are my 3 favorite RPGs (shameless westerner that I am), all have a lot of great jokes in them, even if they are overall quite serious as games... I guess they are just very well written in general. Really, most games I like have some elements of humor in them, even if it's just visual stuff, or stuff suggested by the gameplay... sadistic as it is, the abuse you can put Yoshi through in Super Mario World is pretty funny, and I find the animations in a lot of games to be funny too. Most simulation games, and games where the system is complex enough for their to be emergent behavior of some sort, like Super Smash Bros., can make me laugh too, although I guess it's more out of surprise than because it's funny exactly.

The Ace Attorney series.
Lots of JRPGs make me laugh, but for the wrong reasons. FFX stands out as especially egregious, though that actually made me like the game a lot more than if I had tried to take it seriously.

Wow, so many games have made me laugh they're hard to count. Most have already been mentioned in the thread (the original Sam n Max, SCUMM games, FFVI, etc)
Rather recently, Bayonetta is one of the funniest games I've ever played. I'd also add Elite Beat Agents and the Wario Ware series.
