Why not?
You don't necessarily have to make an FPS. The first Duke Nukem games were platformers. You could also just slap common sense in the face and make a Duke RPG or RTS or whatever.
Or maybe even a Take-Two/3D Realms Sim game where you have to make your employees stop playing WoW and actually make your goddamn game.

This is a splendid idea for a compo. So ah, entries have to be in by sometime around 2021? Now is the only time a DNF compo could really be done. A couple more months and everyone will be over it (except the hard-core fans, and of course the ex-3DRealms employees). We need a list of vaporware games though. Off the top of my mind: Warcraft Adventures DNF Sims Online Advent Rising 2 Fallout Van Buren maybe even Daikatana (the daikatana that was advertised, not the one that was released ) Commander Keen: The Universe is Toast!

OH SHIT! OH SHIT indeed!! I totally forgot about that. That ridiculous teaser in Aliens at my Babysitter was probably my first vaporware experience... And actually, it's the only vaporware game that I'd still be interested in seeing.

By the way, Alan Wake I know it's supposed to come out but it's been years... Also, Tiberium, the C&C FPS.
